Early changes in apoptosis occur on the surface of the cell membrane. One of the changes in the surface of these cell membranes is the transfer of phosphatidylserine (PS) from the cell membrane to the outside of the cell membrane, exposing PS to the outer surface of the cell membrane. PS is a negatively charged phospholipid that normally exists mainly on the inner surface of the cell membrane. When the cell undergoes apoptosis, the asymmetry of the phospholipid distribution on the cell membrane is destroyed and the PS is exposed to the cell membrane. Annexin V has the property of being easily incorporated into phospholipids such as PS and has a high affinity for PS. Therefore, the protein acts as a sensitive probe to detect PS exposed to the surface of the cell membrane. PS transfer to the cell membrane is not unique to apoptosis, but can also occur in cell necrosis. The difference between the two cell death patterns is that the cell membrane is intact at the initial stage of apoptosis, and cell necrosis is destroyed in its early stages. Therefore, early apoptosis of cells can be detected by flow cytometry using Annexin V and PI double staining.
